🧬 Preparing Microscope Slides

Wet Mount Preparation:

  1. Clean slide: Wipe with alcohol or clean cloth
  2. Add specimen: Place small amount in center
  3. Add water/stain: Single drop of mounting medium
  4. Position coverslip: Hold at 45° angle at one edge
  5. Lower slowly: Gradually lower to exclude air bubbles
  6. Remove excess: Blot with filter paper

Common Stains:

  • Iodine solution: Stains starch purple/black, cytoplasm yellow
  • Methylene blue: Stains nuclei dark blue
  • Congo red: Stains cell walls
  • Neutral red: Vital stain, shows living cells

Why stain? Most cells are transparent and colorless. Stains bind to specific structures, making them visible and easier to identify.
